TargetAmp™ aRNA Amplification Kits

TargetAmp™ RNA Amplification Kit Selection Guide

There are currently four TargetAmp™ RNA Amplification Kits available that can be used to prepare labeled target aRNA (cRNA) for use with virtually any DNA microarray platform including Affymetrix GeneChip®, GE CodeLink™ or other commercial chips or self-spotted arrays.

The TargetAmp RNA Amplification Kit selection guide is presented both as a flow-chart and also a table at the end of this page.

In helping determine the best TargetAmp RNA Amplification Kits to meet their needs, the user should first determine:

How much total RNA will I amplify or, how many cells will I extract total RNA from for the RNA amplification process?

Input Total RNA:25 ng - 500 ng total RNA (total RNA from 2500 cells to 50,000 cells)

  • Produces microgram amounts aminoallyl-aRNA for indirect target labeling with a biotin-X-X-NHS, Cy®-NHS or other fluorescent-NHS dye.
  • Greater than 5000-fold amplification of Poly(A) in a total RNA sample.
  • Optimized for use with SuperScript™ III Reverse Transcriptase (provided by the user).
  • Produces microgram amounts unlabeled-aRNA.
  • Greater than 5000-fold amplification.
  • Optimized for use with SuperScript™ III Reverse Transcriptase (provided by the user).
  • Readily modified for direct incorporation of biotin-NTP or fluorescent-NTP (provided by the user).

Target Labeling Options

The TargetAmp 1-Round Aminoallyl-aRNA Amplification Kit 101 produces high yields of aminoallyl-aRNA. Once purified, the aminoallyl-aRNA can be readily labeled with commercially available biotin-X-X-NHS, Cy®-NHS or other fluorescent-NHS dye. There are important advantages to this method of Indirect Target Labeling.

The TargetAmp 1-Round aRNA Amplification Kit 103 produces unlabeled amplified aRNA (cRNA). When using this kit, the final in vitro transcription reaction can be modified for Direct Target Labeling using a biotin-NTP or Cy-NTP or other fluorescent-NTP. Alternately, the unlabeled aRNA produced by the kit can be labeled in a subsequent reverse transcription reaction that includes an aminoallyl-dNTP, a biotin-dNTP, Cy-dNTP or other fluorescent-dNTP.

Input Total RNA: 10 pg - 500 pg total RNA (total RNA from 1 cell to ~50 cells)

  • Produces microgram amounts of Aminoallyl-aRNA for indirect target labeling with a biotin-X-X-NHS, CyR-NHS or other fluorescent-NHS dye.
  • Greater than 5,000,000-fold amplification.
  • Optimized for use with SuperScript™ III & SuperScript™ II Reverse Transcriptases (provided by the user).
  • Produces microgram amounts of unlabeled-aRNA.
  • Greater than 5,000,000-fold amplification.
  • Optimized for use with SuperScript™ III & SuperScript™ II Reverse Transcriptases (provided by the user).

The TargetAmp 2-Round Aminoallyl-aRNA Amplification Kit 1.0 produces high yields of aminoallyl-aRNA. Once purified, the aminoallyl-aRNA can be readily labeled with commercially available biotin-X-X-NHS, Cy®-NHS or other fluorescent-NHS dye. There are important advantages to this method of Indirect Target Labeling.

The TargetAmp 2-Round aRNA Amplification Kit 2.0 produces unlabeled amplified aRNA (cRNA). When using this kit, the final in vitro transcription reaction can be modified for Direct Target Labeling using a biotin-NTP or Cy-NTP or other fluorescent-NTP. Alternately, the unlabeled aRNA produced by the kit can be labeled in a subsequent reverse transcription reaction that includes an aminoallyl-dNTP, a biotin-dNTP, Cy-dNTP or other fluorescent-dNTP.
