Tagetin™ RNA Polymerase Inhibitor

硫黄病を誘発するバクテリアの植物性自然毒として最初に発見された Tagetin™ RNA Polymerase Inhibitor は、様々な真核（生物哺乳動物細胞、Saccharomyces cerevisiae, Drosophila melanogaster, Bombyx mori, Xenopus laevis oocytesなど）のRNA polymerase III を強力にかつ、選択的に阻害する唯一の化合物として知られています¹ 。TargetinはE.coli RNA polymerase やplant chloroplast RNA polymeraseを強く阻害します² 。Targetin は植物RNA polymerases I, II, 及び III には選択性が少なく、SP6やT7ポリメラーゼのようなファージのRNAに対しても比較的選択性はありません³。阻害の程度は、真核細胞及び原核細胞においてもテンプレートに依存します⁴。

Tagetin Inhibitorは、真核細胞のRNA polymerase IIを阻害するα-amanitinを捕捉します。しかしながら、阻害の正確なメカニズムは解明されていません。酵母の核抽出液を用いた研究により、テンプレートの特定の位置でRNA伸長の停止が起こりやすくなることが明らかになっています⁴ 。

Each gel lane shows products of a standard transcription reaction using a bacteriophage template, 1 U of E. coli RNA Polymerase Holoenzyme, and varying amounts of Tagetin Inhibitor. Lane 1, 100 U; Lane 2, 10 U; Lane 3, 1 U; Lane 4, 0.1 U; Lane 5, control without Tagetin Inhibitor. 50% inhibition of transcription is seen at 1 U Tagetin Inhibitor per unit of E.coli RNA Polymerase.

Unit Definition

One unit (30 pmoles) of Tagetin Inhibitor (M.W.=416) results in 50% inhibition of 1 unit of E. coli RNA Polymerase Holoenzyme in 40 mM Tris-HCl (pH 7.5), 150 mM KCl, 10 mM MgCl2, 0.01% Triton X-100, 10 mM DTT, and 0.5 mM each NTP using phage T7 DNA as a template.

Storage Buffer

Tagetin RNA Polymerase Inhibitor is provided in water at a concentration of 20 U/μl (600 μM).

Quality Control

Tagetin Inhibitor is functionally tested with our E. coli RNA Polymerase Holoenzyme and is free of detectable RNase and DNase activities.
