真核細胞のmRNAは、通常immobilized oligo(dT)もしくはoligo(dU)で溶離することで分離されます。mRNA-ONLY™ Eukaryotic mRNA Isolation Kit* は、oligo (dT)に結合させることなしに1時間以内にrRNAを実質的に取り除くシンプルでかつ効果的な方法です。

mRNA-ONLY™ Prokaryotic mRNA Isolation Kit* は、rRNAを取り除いたprokaryotic primary mRNA transcriptsを1時間で得られるシンプルかつ効果的で新しいキットです。真核細胞のpolyadenylated mRNA の精製には、oligo(dT)- やoligo(dU)- immobilized polymersが通常使用されますが、prokaryotic mRNAはポリアデニル化されていないので真核細胞のmRNAにはoligo(dT)は使用できません。

mRNA-ONLYプロトコールは、精製されたtotal RNAから始まります。Total RNAは、MasterPure™ RNA Purification KitやArrayPure™ Nano-Scale RNA Purification Kitなどから精製したものが使用できます。最初のステップは、大きなrRNA (e.g., 18S and 28S)を除去するmRNA-ONLY™ Isolation Solutionによるtotal RNAの処理です。この処理によりmRNAが濃縮されます。キットに付属のLiCl溶液を使用してmRNAを塩沈殿することが可能です。

Applications and Benefits

• Permits quick and easy isolation of eukaryotic and prokaryotic mRNA that is substantially free of contaminating rRNA.
• No oligo(dT) binding step is required, minimizing losses of mRNA due to inefficient binding.
• Simplifies generation of high-quality cDNA libraries or synthesis of specific cDNA.
• Useful for obtaining mRNA for ribonuclease protection assay, RT-PCR, Northern blots, or other procedures.
• Prokaryotic mRNA isolated with mRNA-ONLY can be poly(A)-tailed using the A-Plus™ Poly(A) Polymerase Tailing Kit.
• The mRNA obtained can be amplified using RNA amplification kits, such as TargetAmp™ 1-Round or 2-Round aRNA Amplification Kits, for subsequent gene expression analysis using microarrays or other systems.
• mRNA-ONLY Eukaryotic mRNA Isolation Kit can be used to simultaneously prepare mRNA from both prokaryotic and eukaryotic cells or from mixed populations of prokaryotes, such as occur in biological samples involving symbiotic, parasitic, or pathogenic interactions (e.g., nitrogen fixation, prokaryotic infections, biological films, etc.).
