FosmidMAX™ DNA Purification Kit

FosmidMAX™ DNA Purification Kitは、扱いやすく、また信頼性の高い高品質Fosmid DNAを精製できるキットです。拡張 性のあるプロトコールは一般的に1.5〜100mlのシングル-コピーBAC培養液から0.6〜25 μgのBAC DNAを回収できるアルカリライシス法をベースに修飾したキットです。選択的沈殿ステップおよびEPICENTRE社製RiboShredder™ Rnase Blendの採用で 、分解DNAや、その後のアプリケーションに邪魔となるコンタミを除去します。

Fosmidライブラリーの構築には、CopyControl Fosimid Library Production Kitをお勧めします。独自のCopyControl cloning technologyによりtoxic gene productsやインサートDNAを安定的にsingle-copyでクローニングでき、DNAを得るためにhigh-copyにすることができます。FosmidMAX Kitと組み合わせて使用することで、1.5 mlの培養液から~4μgのfosmid DNAが回収で きます。

Fosmid DNA isolated from 6 different fosmid clones (Lanes 1 to 6) was digested with Hind III and analyzed by agarose gel electrophoresis. Lane M, kb ladder.

DNA from an equal number of cells of uninduced (-) and induced (+) CopyControl fosmid clones was isolated using the FosmidMAX™ Kit. Equivalent volumes of fosmid DNA were digested with Hind III and analyzed by agarose gel electrophoresis. Lane M, kb ladder.

Reagents sufficient for 150 X 1.5 ml; 10 X 40 ml; or 5 X 100 ml purifications
