EZ-Tn5™ Protein Truncation KitとMu-End™ Protein Truncation Kitは、E. coli.で発現可能なN-terminal及びC-terminal protein deletionsのライブラリーを作製する手段です。EZ-Tn5™トランスポゾンは、Transposaseの触媒によるin vitro反応によりどんなターゲットDNAにもランダムに挿入されます。その後、トランスポゾンの末端にアニールするプライマーとターゲット配列にアニールするプライマーでPCRすることにより増幅します。トランスポゾンは、ターゲット配列の長さによってランダムに挿入されるので、プライマーの選択によりPCR増幅の後にはN-terminalもしくは C-terminal deletionsのライブラリーが作製されます(Figure 1)。

キット中のEnd-Repair MixとFast-Link DNA Ligaseを使用することでPCR productのプールはblunt-ended及び self-ligationを行い、kanamycin-resistant "rescue" クローンのライブラリーを作ることができます。p15Aori は標準のE. coli で複製可能です。N-terminal deletionのrescue cloneはT7-promoter 領域を含みます。少なくとも1/3のクローンがT7 RNA polumerase 遺伝子を含む大腸菌(e.g., E. coli BL21)で発現される in-frame protein fusionsを作製できます。

Mu-End™ Protein Truncation Kit は、EZ-Tn5 Protein Truncation Kitで作製されたタンパクのC末端から一方向のdeletionを作製するキットです(Figure 2)。 キットには、56-bp fragment のbacteriophage Muトランスポゾン認識配列R1とR2配列を含みます。in vitro transposition反応によりランダムにMu-endが選択されたクローンに挿入されます。トランスポゾン挿入後、Muに特異的プライマーとEZ-Tn5Transposonに特異的なプライマーでPCRすることでdeletionライブラリーを作製することができます。また、トランスポゾンにより挿入されたoriginやカナマイシンマーカー、T7 promoter 領域はそのまま残ります。Mu トランスポゾン挿入されたDNAはend-repair、ligation反応後に plasmidとして"rescued"することができます。truncated proteinに余分なアミノ酸が加えられるのを避けるために、Muに特異的なプライマーにはストップコドンが含まれています。
